Nassar states in the Preafce that "after 15 years in the field I still cannot find a good reference book to guide the drug metabolism scientist. There are at least two very good books that have filled this purported void already: That said, there is a lot to recommend Al Nassar's book: Chapter One is a good general overview of drug metabolism, setting the stage for implementing strategies that appear later on. Chapter Two is one of the stronger chapters: Chapter Three - really not much new here; the mass shift table is better but is suprisingly listed as "monoisotopic mass changes" when in fact it is a list of exact mass changes Mike Lee's book - mentioned above - excles in this regard.
That said, the glossary that follows Chapter Three is comprehensive and helpful and a good addition. Chapter 4, also by editor Nassar, is a very strong chapter; he describes the medicinal chemistry strategies employed to solve drug metabolism problems. I learned more new from this chapter than any of the others. Chapters 5 and 6 were the most disappointing It is filler and it would have been better to use this space to address some of the shortcomings mentioned in this review.
- Big Winds Horrible Hurricane.
Chapter 7 on Reactive Intermediates was also very strong and comprehensive. Chapter 8 might have been the most relevant chapter as it discusses the latest regulatory guidance on metabolites in safety testing and provides references to some very good case studies. The surprising thing with any of these types of books is that they invariably emphasize that identifying metabolites is becoming an important part of the drug discovery and devlopment process. It should not be a surprise at all I would certainly recommend this book for someone new to the drug metabolism and metabolite ID field as a good overview and compendium of useful references, but there are better books for the nuts and bolts of day-to-day problem-solving for the advanced drug metabolism scientist.

For over five years, Dr. Nassar oversaw the drug metabolism and bioanalytical department at Vion Pharmaceuticals, working in these key areas of drug discovery and development. One principal area of his research involved understanding how structure modification can improve the ADME profile for new chemical entities as they advance toward clinical candidacy. He identified and subsequently patented pharmacologically active metabolites of Nexavar sorafenib , and recently identified the pharmacologically active metabolite of Onrigin laromustine , and is in the process of seeking a patent on this discovery.
